Description

Fluo-Green™ is a modestly water soluble fluoride probe that readily chelates fluoride with high binding constants (Dissociation constant 43 mM3). The probe can be used in either an excitation wavelength ratiometric (colorimetric) or fluorescence emission wavelength ratiometric manner. Optimum emission wavelength ratiometric wavelengths are 450/550 nm, with excitation wavelengths 325-450 nm. The Stern-Volmer fluorescence quenching constants for I-, Br- and Cl- are 34, 1.4 and 1.0 M-1 respectively. The mean and amplitude weighted lifetimes for aqueous (deionized water) Fluo-Green™ are 2.52 and 2.39 ns respectively.

Fluo-Green™ is sold dry and is simply reconstituted by adding water/MeOH. One vial of our fluoride probe will typically allow for 10 fluoride measurements to be undertaken, when further diluted into 3 cm3 volumes within a cuvette. 50 µL of water/MeOH is typically added to each dry vial. Subsequently, ten 5 µL aliquots diluted to 3 cm3, typically give an optical density 0.2 OD.

For optimum performance, probe vials should be stored at -2 to -8 C when not being used. Probe solutions should be used after dilution in deionized water. We recommend the use of Fluo-Green C™, the control compound for Fluo-Green™, to test for background interferences.
